大分子蛋白质激光解吸质谱学研究

摘要 介绍可分析分子量高达100,000道尔顿大分子蛋白质的激光解吸质谱新方法。给出各类蛋白质的激光解吸质谱图例,并论述其特点。使用新方法可以快速(约10min)、准确(分子量测量准确度为0.1~0.2%)、高灵敏度(可分析少至5×10^(-15)mol样品)测定蛋白质及其亚基分子量。还讨论了使用液相基体帮助激光解吸的特点和机理。 A new method of laser desorption mass spectrometry(LDMS)for the analysis of large proteins with molecular weight up to 100,000 daltons is introduced.Laser desorption mass spectra of different kinds of proteins are demonstrated,Their features are discussed.By the use of the new method,the molecular weight of pro- teins and their subunits can be measured fast(within 10 minutes),accurately(with absolute errors of 0.1~0.2%)and with very high sensitivity(amount down to 5 femto-mole proteins is detected).The unique characters and the mechanism of li- quid matrix for assisting laser desorption of large proteins are discussed.
