摘要
目的观察白香丹对经前期综合征(PMS)肝气逆证模型大鼠额区和下丘脑中γ-氨基丁酸(GABA)B1受体(GBR1)、γ-氨基丁酸B2受体(GBR2)分布及表达的影响,探讨白香丹对PMS肝气逆证的干预机制。方法通过旷场实验和阴道涂片法筛选健康雌性Wistar大鼠进入实验,采用电刺激法复制PMS肝气逆证大鼠模型,用免疫荧光双标记技术检测GBR1和GBR2在额区和下丘脑中的表达。结果与正常对照组大鼠比较,模型组大鼠垂直得分、水平得分和总分均显著增加(P<0.01),GBR1和GBR2蛋白在额区和下丘脑中的表达水平降低(P<0.01);与模型组大鼠比较,白香丹组和巴氯芬组大鼠旷场得分显著降低(P<0.01,P<0.05),GBR1和GBR2蛋白在额区和下丘脑中的表达水平明显上升(P<0.01,P<0.05),但两组比较差异无统计学意义(P>0.05)。结论 GABABR亚基(GBR1和GBR2)表达的下调可能与PMS肝气逆证的发生密切相关;白香丹可对GBR1、GBR2的表达起到调节作用。
Objective To investigate the effect of Baixiangdan Capsule on the distribution and expression of γ- aminobutyric acid B1 receptor(GBR1) and γ-aminobutyric acid B2 receptor(GBR2) in the frontal cortex and hy- pothalamus of premenstrual syndrome(PMS) rats with liver-Qi upward invasion syndrome, thus to explore the thera- peutic mechanism of Baixiangdan Capsule for PMS with liver-Qi upward invasion syndrome. Methods After vaginal smear examination and open field test, we chose the healthy female Wistar rats for the experiment, and then built PMS models of liver-Qi upward invasion by electric stimulation. Immune fluorescence double-tagging technology was adopted to detect the protein distribution and expression levels of GBR1 and GBR2 in the regions of frontal cortex and hypothalamus. Results Compared with the normal group, the vertical section score, transverse section score and to- tal score in open field test all increased obviously(P 〈 0.01), and the expression levels of GBR1 and GBR2 signifi- cant decreased in the frontal cortex and hypothalamus(P 〈 0.01 ). Compared with the model group, the scores in open field test decreased significantly (P 〈 0.01, P 〈 0.05) , and the expression levels of GBR1 and GBR2 increased markedly in the frontal cortex and hypothalamus(P 〈 0.01, P 〈 0.05) in Baixiangdan Capsule group and Baclofen group, but there ware no statistical differences between the two treatment groups(P 〉 0.05). Conclusion The lower expression levels of GBR1 and GBR2 in the frontal cortex and hypothalamus may be correlated with PMS with liver-Qi upward invasion syndrome, and Baixiangdan Capsule can regulate the lower expression levels of GBR1 and GBR2.
