摘要
目的:建立高效液相色谱(HPLC)测定鱼血和鱼肝中麻醉剂间氨基苯甲酸乙酯甲烷磺酸盐(MS_222)浓度的方法。方法:样品用重蒸水匀浆,离心(12000r/min)15min,将上清液用0.45μm过滤膜过滤后进YWG_C18柱分析,流动相为0.2mol/L磷酸缓冲液(pH 3.1)∶甲醇=85∶15,紫外检测波长223nm。结果:所得标准曲线的回归方程为鱼血:Y(ng)=0.5258-0.0894X,相关系数γ=0.9939,鱼肝:Y(ng)=-0.100+0.1100X,γ=0.9965,X为峰高。平均回收率为鱼血87.78%、鱼肝76.66%。结论:本法操作简便,重现性和回收率均较好。
Objective:To establish a high performance liquid chromatographic method to determine Finquel(MS-222)in fish blood and hepar.Methods:Samples were homogenized then were centrifuged(1200r/min,15 min),the supernatant retained and microfiltered through 0.45μm micropore membrane prior to injection onto the YWG-C-18,column.The mobile phase was 0.2 mol/L phosphate buffer(pH 3.1):methanol=85∶15,the UV detection wavelength was 223 nm.Results:The calibration curve werey(ng)=0.5258+0.0894X,r=0.9939 for fish blood andY(ng)=-0.100+0.1100X,r=0.9965 for fish hepar,Xwas peak height.The average recoveries were 87.78%for fish blood and 76.66%for fish hepar.Conclusions:This method is simple,rapid and reproducible,and it has a high recovery.
