摘要
用Ficoll Paque和Percoll液 ,使用高速离心法对真鲷末梢血中的淋巴细胞进行分离实验。结果指出 ,用Ficoll Paque液分离时 ,2 1 0 0r/min、2 0℃离心 35min ,活淋巴细胞数占全部被分离白血球的 ( 79.5± 6.5) %。当用Percoll液分离时 ,在 1 40 0r/min、2 5℃离心 30min ,淋巴细胞数占全部被分离白血球的 ( 77.2± 4.3) %。可见 ,这 2种分离培养液都可用于真鲷淋巴细胞的分离 ,但在各自的最佳条件下Ficoll Paque的分离效果略好于Percoll。
There are many reports on methods of separating lymphocytes and leucocytes from peripheral blood of fishes. However, the conditions reported for separation, i.e. density of separation medium, dilution rate of blood, speed for centrifugation etc., are not always applicable to Pagrosomus major (Red Sea Bream). In the experiment as applied to Oncorhynchus kisutch and Japanese ell Anguilla japonica did not show clear results. In this paper, a study was made of was the desirable conditions for separating the lymphocytes of the peripheral blood of the Red Sea Bream by using Ficoll Paque and percoll. The results obtained from Ficoll Paque and Percoll separations showed that Ficoll Paque separation resulted 79% (S.D. 6.5) purity of lymphocyte whereas Percoll exporation resulted in 77.2% (S.D. 4.3) purity. Therefore the seperation of lymphocytes using Ficoll Paque fluid is better than that using Percoll separation.
