摘要
【目的】用生物信息学方法探讨糖尿病相关基因KCNJ11的结构及其蛋白质的结构和功能。【方法】利用ORF Finder、PSIPRED、ProtScale、Swiss Model、CKSAAP_GlySite等在线服务器以及ClustalX和Pymol等工具对KCNJ11基因结构以及蛋白质的基本性质包括亲疏水性、蛋白质翻译后修饰、二级结构和三维结构进行分析。【结果】亲水性氨基酸平均分布在KCNJ11中,同时,它还富含无规则卷曲二级结构原件。通过预测软件发现它共发生两种蛋白质翻译,包括磷酸化和糖基化。将KCNJ11与它的同源蛋白质进行比对,发现它含有8个关键氨基酸。【结论】通过对KCNJ11基因及其产物蛋白质的生物信息学分析,对它的结构和功能有了更进一步的了解。
[Objective] To investigate the structure and function of Type 2 diabetes related gene KCNJ11 by bioinformaties methods. [Method] The KCNJll gene structure and its physical properties have been obtained from NCBI database. Meanwhile, the hydrophilicity scales, post-translation modifications, secondary structure and three dimension strueture of KCNJ11 expression product protein have also been investigated with online tools PSIPRED, ProtScale, Swiss Model and CKSAAP_GIySite as well as the softwares ClustalX and Pymol. [ Result] Hydrophilic amino acids are evenly distributed across the KCNJ11 protein sequence. Protein secondary structure contains et-helix, 13-sheet, [3-turn and random coil, among which the random coil was frequently appeared in KCNJ11 protein structure. There were two kinds of protein post-translation modifications occurred in KCNJ11 protein, including glycosylation and phosphorylation. In addition, eight key amino acids of KCNJll have been identified by alignment with its homology protein. [Conclusion]The structure and function of gene KCNJll and its expression product protein have been intensively investigated by bioinformatics analysis.
