摘要 试验旨在建立内蒙古绒山羊皮肤蛋白质双向电泳图谱。采用液氮研磨法提取蛋白质,通过双向电泳技术对蛋白质进行分离,分别对不同的蛋白质沉淀方法、上样量和SDS-PAGE凝胶浓度进行比较。结果显示,使用2-D Clean-up试剂盒处理、上样量为20μg、采用10%的SDS-PAGE凝胶浓度时可获得重复性好和分辨率高的双向电泳图谱,便于后续的内蒙古绒山羊皮肤差异蛋白质组学分析。 This study was aim to establish an efficient and high resolution proteomic map of skin collected from Inner Mon- golia Cashmere goat. Total protein was extracted by using liquid nitrogen grinding method. The effects of different precipitate methods, different loading quantities and different gel concentration of SDS-PAGE on 2-DE (two-dimensional gel electrophore- sis) maps were compared and optimized though the 2-DE. The results showed that the best electrophoresis pattern could be ob- tained by using 2 D Clean-up kit in protein extracting and loading 20 μg protein samples on each strip as well as using SDS- PAGE with 10% gel concentration and that would be helpful for the further proteomics analysis.
