骨桥蛋白靶向干扰RNA对体外人脐静脉内皮细胞增殖和凋亡的影响

摘要 目的:观测骨桥蛋白靶向干扰RNA对体外人脐静脉内皮细胞(HUVEC)增殖和凋亡的影响。方法:体外化学合成骨桥蛋白序列特异性的短发夹RNA(shRNA)质粒载体与阳离子脂质体结合介导转染HUVEC,同时设计空白载体转染对照和空白对照,用MTT法等方法检测各组HUVEC生长增殖情况的变化,并用流式细胞术检测细胞凋亡情况。结果:OPN-shRNA载体转染HUVEC后明显抑制细胞增殖(P<0.05)和使细胞分裂周期出现阻滞,凋亡二倍体数量增加,凋亡率显著升高(P<0.05)。结论:骨桥蛋白靶向干扰RNA在体外能够抑制HUVEC的增殖,升高细胞凋亡率。
