摘要
目的 :观察缓慢牵伸肢体延长时周围神经相应的脊髓及背根神经节(DRG)中生长相关蛋白 43(GAP 43)mRNA表达。方法 :29只日本大耳白兔胫骨以1mm/d的速度缓慢牵伸延长至20 %、40 %,停止延长后2、4周 ,用原位杂交技术观察坐骨神经相应背根神经节及脊髓中GAP 43mRNA的表达及定位 ,以图像分析仪进行半定量分析。结果 :缓慢牵伸至20 %时相应脊髓及神经节呈阳性表达 ,延长40 %时呈强阳性表达 ,停止延长4周恢复正常。结论 :缓慢牵伸周围神经虽无神经临床损害症状 ,但引起GAP 43mRNA表达的上调 ,并在停止延长后4周恢复正常 ;缓慢牵伸过程中神经的亚临床损害广泛存在 ,并存在着强烈的神经再生反应。
Objective: To investigate the expression of GAP 43 mRNA in corresponding DRG and spinal cord during and after the limb lengthening. Methods: Using in situ hybridization, we investigated the expression of GAP 43 mRNA in corresponding spinal cord and dorsal root ganglion after 20% and 40% lengthening and at the 2nd and 4th week after 40% lengthening in rabbits. Results: There was positive expression in 20% lengthening group and stronger expression in 40% lengthening group. But the expression recovered to normal at the 4th week after limb lengthening. Conclusion: The expression of GAP 43 mRNA can be up-regulated during limb lengthening, which is corresponded to the subclinical damage of peripheral nerve. However, it can gradually recover to normal after the lengthening stops.
