TRAIL及其受体在肿瘤治疗中作用机制研究的现状与展望 被引量:3

摘要 肿瘤坏死因子相关凋亡诱导配体(TNF related apoptosis inducing ligand,TRAIL) 是肿瘤坏死因子(tumor necrosis factor,TNF)超家族中的一员,最早由Wiley等[1] 从人心肌cDNA文库中克隆出来并命名.肿瘤坏死因子家族(tumo r necrosis factor,TNF)有3种分子可诱导凋亡,TNF、FasL和TRAIL.TRAIL与前两者比较,不同之处在于,具有强大和特异性诱导肿瘤细胞凋亡的能力,但对正常细胞无此效应;有更广的抗癌谱;对核因子-κB(nuclear factor-κB,NF-κB)仅有很微弱的激活作用,即使是全身用药,也不会产生严重的炎症反应.因此,TRAIL 被认为是一种更为安全且极具潜力的抗肿瘤因子.本文就其结构功能及抗肿瘤机制研究最新进展综述如下.
