摘要 目的:研究粘着斑激酶(focal adhesion kinase,FAK)对舌鳞癌细胞系Tca8113细胞侵袭和迁移的影响。方法:用RNA干扰的方法抑制Tca8113细胞FAK基因的表达。然后以划痕试验测定细胞迁移,以Transwell小室侵袭模型研究细胞侵袭能力。结果:外源性干扰质粒可以显著抑制Tca8113中FAK的表达;FAK基因沉默后,Tca8113细胞迁移能力、侵袭能力明显下降。结论:FAK在舌鳞癌Tca8113细胞的迁移和侵袭过程中发挥重要作用。 Objective: To investigate the effects of focal adhesion kinase(FAK) on the invasion and migration of human oral squamous cell carcinoma Tea8113 cells. Methods: RNA interference(RNAi) method with pSilencer-FAK vector was used to inhibite FAK expression. The velocity of cell migration was measured by wound-repair experiment. The invasion of the cells was detected by invasion chamber assay. Results : FAK expression of the RNAi treated ceils was suppressed by pSilencerFAK vector. After FAK gene silencing, the velocity of migration and invasion of the cells were inhibited significantly. Conclusion : FAK can promote migration and invasion of Tea8113 ceils.
