不同十八碳脂肪酸对奶牛乳腺上皮细胞增殖及甘油三酯合成的影响 被引量:5

Effects of 18-carbon Fatty Acids on Cell Proliferation and Triacylglycerol Accumulation in Bovine Mammary Epithelial Cells in vitro

摘要 本研究旨在探讨外源添加十八碳脂肪酸对体外培养的泌乳奶牛乳腺上皮细胞增殖及甘油三酯合成的影响。以体外培养泌乳奶牛乳腺上皮细胞为模型,添加不同浓度的硬脂酸、油酸、亚油酸及亚麻酸于38℃培养24h;采用MTT法检测细胞增殖情况,采用甘油三酯检测试剂盒检测培养基中甘油三酯的含量。结果,与对照相比,200、400μmol.L-1的硬脂酸、油酸、亚油酸和亚麻酸均能显著或极显著抑制细胞的增殖(P<0.05或P<0.01);甘油三酯的检测结果显示,在0~100μmol.L-1的添加范围内,4种十八碳脂肪酸均能显著或极显著的增加培养基中甘油三酯的含量(P<0.05或P<0.01),且培养基中甘油三酯的含量随各十八碳脂肪酸添加量的增大而增大。结果提示,较高浓度(200、400μmol.L-1)的硬脂酸、油酸、亚油酸和亚麻酸能抑制细胞增殖;而在泌乳奶牛乳腺上皮细胞生长增殖不受抑制的条件下(脂肪酸添加浓度为0~100μmol.L-1),甘油三酯的合成与各十八碳脂肪酸添加剂量呈正相关。 The experiment was conducted to study the effects of 18-carbon fatty acids(18C-FAs) on cell proliferation and triacylglycerol accumulation in bovine mammary epithelial cells cultured in vitro.The cells were cultured with different concentration of stearic acid,oleic acid,linoleic acid and linolenic acid for 24 h at 38 ℃,and then the MTT experiment was performed to detect the cell proliferation and the triacylglycerol contents in medium were detected by Triglyceride Quantitation Kit.The results showed that the cell proliferation was inhibited significantly when the cells were cultured with 200 or 400 μmol·L-1 stearic acid,oleic acid,linoleic acid or linolenic acid(P〈0.05 or P〈0.01).The contents of triacylglycerol in medium were increased in a concentration-dependent manner from 0 to 100 μmol·L-1(P〈0.05 or P〈0.01).In conclusion,high concentration of 18C-FAs can inhibit the cell proliferation,and triacylglycerol accumulation can be increased as the concentration increase of 18C-FAs.
