RP-HPLC法测定“加味白头翁”颗粒中盐酸小檗碱的含量 被引量:3

摘要 为了建立"加味白头翁"颗粒剂中盐酸小檗碱含量的反向高效液相层析(reverse phasehigh-performance liquid chrom atography,RP-HPLC)法。采用KromasilC18柱(4.6mm×250mm,5μm),流动相:乙腈与0.03mol/L磷酸二氢钾溶液体积比30:70的混合溶液,流速1mL/min,紫外检测波长:265nm,柱温:30℃。在所选的色谱条件下,盐酸小檗碱峰与辅料、溶剂及复方提取物中的其他成分的峰分离良好,盐酸小檗碱在93.00~186.00μg/mL范围内呈良好的线性关系(r=0.9997,n=6),平均回收率为99.49%。RSD=0.76%。所用方法简便、灵敏,可用于"加味白头翁"颗粒剂的质量控制。 To establish a simple RP-HPLC method for determination the content of terberine hydrochloride in Modified Pulsatilla granule. HPLC method was used. The HPLC system consisted of Ture Kromasil C18 column (4.6 min×250 ram, 5μm) and acetonitrile-0.03 mol/L phosphoric acid (30:70, r v) mixture as the mobile phase, with a flow rate of 1.0 mL/min. The detection wavelength was 265 nm. The column temperature was maintained at 30℃. The excipients and solvents in the granule could be well separated from the drug under such a designated chromatogram condition and did not interfere with the assay. A good linear relationship was found between peak area and the concentration of berberine hydrochloride in the range of 93.00-186.00μg/mL (r=- 0.9997, n=6). The average recovery was 99.49%, and RSD was 0.76% (n=6). The established approach was specific, accurate, reliable, prompt, sensitive and applicable. It could be used to control the quality of Modified Pulsatilla granule.
