摘要
本研究测定了NIDDM病人(n=6)和正常人(n=7)循环血中单核细胞(Mo)结合^(125)I-胰岛素总能力、胰岛素-受体复合物进入能力、胰岛素降解能力和趋溶酶体制剂氯喹对受体后过程的影响。结果显示NIDDM时Mo对胰岛素的结合能力、进入能力和降解能力均明显低于正常人。这些改变可能与NIDDM胰岛素抵抗性有关。
We studied the total cell-associated A14-125I-insulin radioactivity (TBd, including surface-bound and internalized radioactivity), insulin internalization (In), and the effects of lysosomotropic agent Chloroquine (CQ) on postreceptor processing in peripheral monocytes (Mo) from the patients with non-insulin-dependent diabetes mellitus (NIDDM, n=6) and normal controls (n= 7). The results indicate that 1) insulin is able to bind to and internalize into Mo; 2) the abilities of TBd and In of Mo from the patients with NIDDM are significantly decreased (4.03±0.40 vs 9.40±0.87 and 1.95±0.44 vs 6.06±0.63 pg/107Mo); 3) intracellular insulin degradation is reduced in Mo from the diabetics; and 4) lysosomotropic agent CQ, has no effects on intracellular A14-125I-insulin radioactivity in Mo from both diabetic and normal subjects. These defects may be related to the cellular insulin resistance present in patients with NIDDM.
