皖南花猪骨骼肌AdpR和MyHC mRNA表达的发育性变化 被引量:1

Developmental patterns of adiponectin receptors and myosin heavy-chain isoform mRNA expressions in skeletal muscle of Wannanhua pigs

摘要 为探讨皖南花猪骨骼肌组织中脂联素受体(AdpR1、AdpR2)和不同类型肌球蛋白重链(MyHC)mRNA的发育性变化及性别差异,选择0(出生当天)、30、45、90、180日龄的皖南花猪公母各5头,以β-Actin为内标,采用△△Ct相对定量实时荧光PCR方法对背最长肌和半腱肌中AdpR1、AdpR2、MyHCI、MyHC2a、MyHC2b和My-HC2xmRNA进行定量分析。结果显示,背最长肌和半腱肌AdpR1、AdpR2、MyHC1、MyHC2a、MyHC2b和My-HC2xmRNA的表达都有显著或极显著的发育变化规律(P<0.05或P<0.01)。总体上AdpR1、AdpR2、My-HC2a、MyHC2b和MyHC2xmRNA在背最长肌显著或极显著高于半腱肌(P<0.05或P<0.01);MyHC1mRNA在背最长肌极显著低于半腱肌(P<0.01)。半腱肌MyHC1mRNA在母猪显著大于公猪(P<0.05),而MyHC2amRNA在母猪显著小于公猪(P<0.05)。背最长肌和半腱肌AdpR2mRNA的表达分别与MyHC1正相关(P<0.05),半腱肌AdpR1mRNA的表达与MyHC2x正相关(P<0.05)。结果表明,皖南花猪骨骼肌组织中AdpR和MyHC的基因表达有特定的发育模式和组织特异性,且有一定性别差异。 Adiponectin(Adp) is a cytokine secreted mainly by adipose tissue,which has important physiological func- tions. The objective of this study was to explore the developmental patterns and sexual dimorphism of adiponectin re- ceptors (AdpR1, AdpR2), myosin heavy chain (MyHC) isoform mRNA expression in skeletal muscle of Wannanhua pigs. Five Wannanhua boars and five Wannanhua gilts were sampled at bitrth, 30,45,90, and 180 days of age respec- tively. The AACt relative quantification real-time PCR was used to detect the transcription levels of AdpR1, Ad- pR2, MyHC1, MyHC2a, MyHC2b and MyHC2x mRNAs in longissimus dorsi (LD) and semitendinosus (ST), and - actin were used as internal standards. The expression level of AdpR1, AdpR2, MyHC1, MyHC2a, MyHC2h and My- HC2x mRNA in LD and ST were changed with age significantly(P%0.05 or P%0.01). In general,AdpR1 ,AdpR2, MyHC2a,MyHC2b and MyHC2x mRNA expression in LD was significantly higher than those in ST (P%0.05 or P %0.01) ,while MyHC1 mRNA expression in LD was significantly lower than those in ST(P〈0.01). The expres- sion of MyHC1 mRNA in ST of Wannanhua gilts was significantly higher than those in boars, but MyHC2a mRNA expression in ST of Wannanhua gilts were significantly lower than those in boars in general. Although the sexual di- morphism was found in apart genes or apart days of age,AdpR1, AdpR2, MyHC2b and MyHC2x mRNA expression both in LD and ST had no significant differences between Wannanhua gilts and boars in general. Significant positive correlation was found between AdpR2 and MyHC1 (P%0.05) in LD and ST respectively,and significant positive correlation was found between AdpR1 and MyHC2x (P%0.05) in ST. The expression of AdpR and MyHC mRNA in skeletal muscle of Wannanhua pigs followed temporal and spatial specificity, and showed gender dimorphism to some extent.
