β-七叶皂苷钠对大鼠局灶性脑缺血再灌注后AQP4蛋白表达的影响 被引量:7

摘要 目的探讨大鼠局灶性脑缺血再灌注脑组织缺血区AQP4蛋白的表达情况及β-七叶皂苷钠的治疗作用和可能机制。方法采用大鼠大脑中动脉闭塞法(MCAO)制作局灶性脑缺血再灌注模型。应用干-湿比重法、TTC染色、免疫组化SABC法分别测定脑含水量、脑梗死体积及水孔蛋白4(aquaporin4,AQP4)的表达。结果假手术组AQP4蛋白有轻微的表达,而模型组、治疗组缺血损伤后表达升高,脑含水量增高,梗死脑组织出现,给予β-七叶皂苷钠的治疗组AQP4蛋白的表达下降,脑含水量明显降低,脑梗死体积减少,且β-七叶皂苷钠中高剂量组明显优于低剂量组(P<0.05)。结论脑缺血再灌注脑组织缺血区AQP4蛋白表达的上调与脑水肿的发生发展与有密切的关系,而β-七叶皂苷钠有抑制脑水肿发生的作用。 Objective To invstigate the expression of AQP4 in rat brain after transient focal cerebral ischemia/ reperfusion injury,to observe the therapeutic effects and to explore the mechanisms of β-sodium aescinate. Methods Rat models of transient focal cerebral ischemia/reperfusion was set up by using the method of middle cerebral artery occlusion ( MCAO ), the brain water content was measured by using dry-wet weight assay,the brain infarct volume was detected by using the technique of TYC, the expressions of AQP4 in rat brain were detected by immunohistochemical SABC method. Results There was a slight expression of AQP4 in sham-operated group with no significant difference compared to contra- lateral one, AQP4 protein expression in model group and treatment group was significantly increased, the brain water content increased and the brain infarct volume appeared,the expressions of AQP4 in rat brain and brain water content were signifi- cantly decreased for the treatment groups by giving β-odium Aescinate, and the brain infarct volume was reduced too. The effect of aescin in the middle-high dose group was superior to low-dose group (P 〈 0.05 ). Conclusion The occurrence and development of cerebral edema is closely related to the increase in AQP4 protein expression. In cerebral ischemic terri- tory,β-sodium aescinate has the function of reducing brain edema.
