摘要
电子克隆并探讨了一个烟草Mlo基因的结构和功能.以GenBank公布的水稻Mlo基因序列为信息探针对烟草的EST数据库进行同源搜索,并将获得的同源性高的EST序列进行序列拼接,最后得到了一个烟草Mlo基因的cDNA序列.采用生物信息学方法分析该基因编码蛋白的一级、二级结构,并对其功能进行预测.结果表明,此烟草Mlo基因编码一个由288个氨基酸组成的蛋白,相对分子质量为33kD,有7个跨膜区域,蛋白二级结构以α螺旋为主,有1个DUF1084超家族的保守结构域.蛋白功能分析表明,该烟草Mlo基因编码蛋白具有跨膜介导信号传递的功能,可能是一种类似于G蛋白偶联受体的膜结合转运蛋白.
To understand the structure and functions of the tobacco Mlo gene, it was cloned in silicon by bioinformatics tools. The tobacco EST database was searched with the rice Mlo gene (GenBank accession number. Os03g36790.1) served as a pmbe;a cDNA sequence of a tobacco Mlo gene was obtained by assembling the ESTs with high homology. The first-level and second-level structure as well as function of the protein encoded by the gene were analyzed by bioinformafics. The findings show that the gene of tobacco Mlo encoded a protein which is composed of 288 amino acids and the relative molecular mass is 33 kD with seven transmembrane domains. The secondary structure of the protein mainly consists of alpha helixs, with a conservative superfamily structure domain DUF1084. It is concluded that protein function analysis suggests that the protein encoded by the tobacco Mlo gene has the function of transmembrane signal transduction, which works as a protein which is similar to the membrane-bound transporter protein of G-protein coupled receptor.
