加巴喷丁和米诺环素对癫癎持续状态大鼠海马N-甲基-D-天冬氨酸受体表达的影响 被引量:2

Effects of Gabapentin and Minocycline on Expression of N-Methyl-D-Aspartate Receptor Subunit 1 in Hippocampal Neurons of Status Epilepticus Rats

摘要 目的研究加巴喷丁(GBP)和米诺环素对戊四氮(PTZ)致癫癎持续状态(SE)大鼠海马区N-甲基-D-天冬氨酸受体(NMDAR1)表达的影响。方法 SD大鼠84只随机分为对照组、PTZ致组(PTZ组)、GBP 600 mg组(GBP组)、GBP 600 mg干预组(干预组1)、米诺环素45 mg干预组(干预组2)、米诺环素90 mg干预组(干预组3)、米诺环素180 mg干预组(干预组4),每组12只。采用PTZ溶液60 mg.kg-1.d-1腹腔注射诱发SE大鼠,PTZ注射期间出现RacineⅣ~Ⅴ级的惊厥发作持续30 min,连续3 d未死亡者为SE点燃成功。GBP组予GBP灌胃,对照组给予9 g.L-1盐水灌胃。采用头皮针状电极单极导联法观察大鼠性放电的脑电图。免疫组织化学染色检测NMDAR1表达。结果 NMDAR1表达在PTZ组显著高于对照组(P<0.01);GBP组与对照组比较差异无统计学意义(P>0.05);干预组1表达低于PTZ组(P<0.05)。3个不同剂量的米诺环素组NMDAR1表达与PTZ组比较差异均无统计学意义(Pa>0.05)。结论 PTZ致SE大鼠海马神经元NMDAR1表达增加,GBP能降低PTZ所致的SE大鼠NMDAR1表达增加,而米诺环素45~180 mg.kg-1不能降低PTZ所致的SE大鼠NMDAR1表达增加。 Objective To investigate effects of gabapentin(GBP) and minocycline(MINO) on expression of N-methyl-D-aspartate(NMDA) receptor subunit 1(NMDAR1) in hippocampus of status epilepticus(SE) rats induced by pentylenetetrazol(PTZ). Methods Eighty-four SD rats were randomly divided into 7 groups(12 each group): control group,PTZ group,GBP group(600 mg·kg-1·d-1),GBP treatment group(600 mg·kg-1·d-1) and 3 MINO treatment groups(45 mg·kg-1·d-1,90 mg·kg-1·d-1,180 mg·kg-1·d-1).SE model was induced by PTZ in rats.PTZ(60 mg·kg-1·d-1) was injected into the abdominal cavity of rats in PTZ and treatment groups for 3 days. GBP was given to GBP group and saline was given to control group.Epileptic discharge was observed in rat electroencephalogram(EEG) by needle electrodes with unipolar leads in the scalp.The expression of NMDAR1 was detected by immunohistochemistry. Results The expression level of NMDAR1 in PTZ group was significantly higher than that in control group(P〈0.01),while no significant difference between GBP group and control group(P〉0.05).The expression level of NMDAR1 in GBP treatment group was significantly lo-wer than that in PTZ group(P〈0.05),while no significant difference was found in 3 MINO treatment groups and in PTZ group(Pa〉0.05). Conclusions PTZ increases the expression of NMDAR1 in hippocampus neurons of SE rats.GBP can decrease the expression of NMDAR1 in hippocampus neurons of SE rats,while MINO(45-180 mg·kg-1·d-1) can′t decrease the expression of NMDAR1 in SE rats induced by PTZ.
