摘要
目的研究槟榔碱对钉螺足跖平滑肌影响及其机制。方法采用离体钉螺足跖平滑肌收缩活动的实验方法和全细胞膜片钳技术,探讨槟榔碱的作用特点和对Ca^(2+)通道的影响。结果3μmol/L槟榔碱可增加足跖平滑肌的收缩力和收缩频率。 30 nmol/L~1μmol/L摈榔碱直接开放豚鼠单个心室肌细胞钙通道,使I~V曲线峰值增大,激活曲线左移。结论较低浓度的槟榔碱可增加钉螺足跖平滑肌的收缩活动,此作用可能与该药直接开放该通道,促使Ca^(2+)内流有关。
Objective To study the effect of arecoline(Are) on contracted activation and its mechanism. Methods Isolated metatarsus smooth muscles from Oncomelania were used to study the effect of Are on contraction. Patch clamp whole cell recording techniques were used to examine the effect of Are on calcium currents in single guinea pig ventricular myocyte. Results 3 μmol/L of Are obviously increased the contraction activities of smooth muscles. 30 nmol/L-1 μmol/L of Are increased the peak amplitude of Ca^(2+) current in I - V relation curves, and the activation curves shifted to left. Conclusions Relatively lower concentration of Are significantly enhanced the contraction activation in mettarsus smooth muscles of Oncomelania. Furthermore, it suggested that this effect was related to I_Ca activated directly.
