摘要
建立了用丹磺酰氯定性鉴定吡咯喹啉醌的薄层色谱法。甲基营养菌No.2培养液经二乙氨基乙基交联葡聚糖凝胶A -25离子交换剂、Sep -PakC18 Cartridge纯化后 ,可获得纯度92 %以上的吡咯喹啉醌 (PQQ)粉末。在碱性条件下丹磺酰氯 (DNS_Cl)可与PQQ生成具有特征性的DNS_N_PQQ荧光衍生物 ,将其点样于聚酰胺薄膜上 ,经正庚烷 -正丁醇 -醋酸 (体积比3∶3∶1)系统展开 ,Rf 值为0.73。
A thin layer chromatographic method for the qualitative analysis of pyrroloquinoline quinone(PQQ) with dansyl chloride(DNS _ Cl) is reported. PQQ powder with purity above 92% can be obtained from the inoculum of methylotrophic bacteria No.2 purified by DEAE _ Sephadex A-25 and Sep-Pak C18 Cartridge. Under basic conditions PQQ reacts with DNS _ Cl to form a characteristic fluorenscence substance DNS _ N _ PQQ. The sample was developed on polyamide sheet by using n _ heptane-n _ butanol-acetic acid(3∶3∶1 by volume)system as developing solvent, and a fluorescence spot with Rf=0.73 was obtained.
