摘要
目的 :探讨三氧化二砷 (As2 O3)对结肠癌细胞的凋亡诱导作用及其与细胞周期的关系。方法 :采用透射电镜、丫啶橙荧光染色、TdT及流式细胞仪检测药物作用前后细胞在形态学及细胞周期分布等方面的变化。结果 :形态学观察发现As2 O3 诱导的LoVo细胞死亡呈现凋亡特征 ,研究表明随着药物作用时间延长 ,凋亡细胞比例逐渐增加。As2 O3 在低浓度时主要干扰细胞在S期的通过 ,高浓度时则选择性诱导S期细胞凋亡。结论 :As2 O3 可能是通过诱导细胞凋亡而抑制LoVo细胞增殖 ,这种杀伤作用存在周期特异性。对此研究有助于治疗方案的合理设计。
Purpose:To evaluate the effect of apoptosis induced by Arsenic trioxide (As 2O 3) on colon cancer cell line (LoVo) and its relations to cell cycle. Methods:LoVo cell cycle phase distribution was measured by EM, acridine dying、TdT and FCM. Results:Morphological observation indicate that As 2O 3 induced LoVo cell death characterized by apoptotic phenotype. Efficiency of apoptosis was enhanced in time dependent pattern. Lower concentrations of As 2O 3 pertub cells progressing through S/G2 phase, while higher concentrations induce S phase cell apoptosis selectively. Conclusions:Induction of apoptosis contribute to the mechanism for the inhibitory effect of As 2O 3 and this killing effect has shown cell cycle specificity. This knowledge may be helpful designing an approprite chemotherapeutic regimen.
