摘要
目的 :运用高效毛细管电泳 (HPCE)分离 6种蛋白质。方法 :以线性高分子聚合物聚乙二醇 (PEG2 0 0 0 0 )作为筛分介质 ,15 0mmol/LTris 10 0mmol/LCHES(pH8.7)作为运行缓冲液 ,运行电压 2 0kV。结果 :磷酸化酶B、牛血清白蛋白、肌动蛋白、碳酸酐酶、烟草花叶病毒外壳蛋白和胰蛋白酶抑制剂达到基线分离。结论 :高效毛细管电泳分离生物大分子具有快速。
Aim:To separate proteins by high performance capillary electrophoresis(HPCE). Methods:The method of polymethlene glycol (PEG20000) used as separation matrix was developed for the separation of proteins. The running buffer was composed of 150 mmol/L Tris 100 mmol/L CHES and the running voltage was 20 kV. Results: Proteins which were phosphrylase (MW 94000),serum albumin(MW 67000), actin(MW 43000),carborin anlydrase(MW 30000),Tobacco Mosaic virus shell protein(MW 17500) and trypsin inhibitor(MW 20100)were resolved successfully. Conclusion: HPCE with the characteristics of rapidity, high performance is useful to separate proteins.
