腺病毒载体介导IL-24基因对肺癌细胞的化疗增敏效应 被引量:4

摘要 目的研究携带人IL-24基因腺病毒载体(Ad-IL-24)联用顺铂(DDP)对人非小细胞肺癌NCI-H460细胞生长和凋亡的影响。方法取对数生长期的NCI-H460细胞,MTT比色法检测Ad-IL-24联合DDP对细胞生长的影响,流式细胞仪分析细胞周期及检测细胞凋亡率。结果 Ad-IL-24联合DDP对NCI-H460细胞的生长抑制作用和诱导凋亡效应均明显优于单用Ad-IL-24和单用化疗药物DDP,且G2/M期细胞比例显著增加(P<0.05或<0.01)。结论 Ad-IL-24能提高NCI-H460肺癌细胞对化疗药物DDP的敏感性。 Objective To investigate the effects of recombinant adenovirus expressing IL-24 (Ad-IL- 24) combined with cisplatin(DDP) on growth and apoptosis of human non-small cell lung cancer NCI-H460 cells. Methods NCI-H460 cells in logarithmic phase were prepared. Effects of Ad-IL-24 combined with DDP on cell growth of NCI-H460 cells were detected by MTI" assay. Cell cycle and apoptosis were analyzed by flow cytometry. Results Compared with any single group, the inhibition of cell gro^h and cell apoptosis in com- bined group are more effective. Furthermore, Ad-IL-24 combined with DDP significantly increased the ratio of cells in G2/M phase(P 〈0.05 or 〈0.01). Conclusion Recombinant adenovirus expressing IL-24 could en- hance the sensitivity of lung cancer cells to chemotherapeutic drugs DDP.
