摘要
目的优选黄芩苷的水煎煮提取工艺,建立黄芩苷的含量测定方法。方法通过正交实验设计,以黄芩苷总量为考查指标,优选最佳提取工艺;用HPLC法测定黄芩苷含量。结果最佳水煎煮工艺为12倍药材量水煎煮,3次,每次1h。在0.046 52~0.465 2μg范围内线性关系良好,r=0.999 9,(n=6);平均加样回收率为99.98%,RSD=1.56%(n=9)。结论该提取工艺合理、稳定、可行;含量测定方法准确、精确、重复性好,适用于黄苓苷提取物的含量控制。
Objective To establish an appropriate extracting technology for Scutellariae Radix extracts by wa- ter decoction extraction method, and establish a method for baicalin content determination. Methods The total a- mount of baicalin in Scutellariae Radix extracts was used as the index by orthogonal design to optimize the best technique. HPLC was adopted for the determination of baicalin in Scutellariae Radix extracts. Results The opti- mized parameter resulted when the water consumption was 12 times the weight of Scutellariae Radix, which was de- cocted three times, once per hour. A linearity was obtained from the range of 0. 046 52 -0. 465 2μg; with a good correlation coefficient ( r = 0. 999 9, n = 6 ). The average recovery was 99.98 %, RSD = 1.56% ( n = 9 ). Conclu- sion The extraction technique was appropriate for Scutellariae Radix extracts; the technique was stable , rational and feasible. The HPLC method was accurate, precise, and repeatable in estimating the contents of baicalin in Scutellariae Radix extracts.
