Effect of embryonated egg extracts on splenocyte and macrophage in vitro

摘要 目的:以体外细胞实验研究鸡胚蛋提取物对免疫力低下小鼠脾淋巴细胞和巨噬细胞的影响。方法:以水和石油醚为提取介质,检测鸡胚水溶物(CEAE)和脂溶物(CEPEE)在10~500μg/mL浓度范围内对脾淋巴细胞增殖、脾淋巴细胞分泌IL-2、巨噬细胞吞噬作用以及巨噬细胞产NO的影响。结果:鸡胚蛋提取物能恢复免疫低下小鼠的免疫功能,CEPEE显著地促进脾淋巴细胞增殖并分泌IL-2,而CEAE则对巨噬细胞的吞噬能力和分泌NO能力显示出更强的刺激作用,两种提取物均呈现出量效关系。结论:鸡胚蛋脂溶物和水溶物分别作用于淋巴细胞和巨噬细胞,发挥正向免疫调节作用。 Objective:To study the effect of embryonated egg extracts on splenocyte and macrophage of hypoimmunity mice by in vitro cell trials.Methods:Hydro-ligarine was applied as the extraction solvent to divide embryonated egg mixture to hydro-soluble(CEAE)and petroleum-ether-soluble(CEPEE)part.The investigation of immunomodulatory effect of CEAE and CEPEE consisted of splenocyte proliferation,IL-2 production,macrophage phagocytosis and NO production at concentration of 10~500μg/mL.Results:Embryonated egg extracts recovered the the immune function.CEPEE remarkably enhanced splenocyte proliferation and IL-2 production,while CEAE significantly contributed to the macrophage function(macrophage phagocytosis and NO production),both in a dose-dependent way.Conclusion:The hydro-soluble and petroleum-ether-soluble parts acted on the splenocyte and macrophage respectively to up-regulate cell immunity.
