产原果胶酶菌株的筛选及其产酶条件优化 被引量:2

Screening of strain producing protopectinase and optimizing the fermentation conditions

摘要 从土壤中筛选得到1株高产原果胶酶的菌株WJ-2,经初步鉴定为曲霉属(Aspergillus sp)。研究了菌种在不同初始pH值、温度、培养时间、接种量下的产酶情况并进行了产酶条件优化。通过单因素实验和正交实验确定了该菌株液体产酶的最佳条件:初始pH5.0、温度35℃、培养时间48h、接种量为10%,优化条件下的原果胶酶活力可达4545.235U/mL。 One strain W J-2 from soil with high protopectinase yield was screened and initially identified as Aspergillus (Aspergillus sp). The conditions for protopectinase production of the strain were studied and optimized at different initial pH value, temperature, culture time, inoculation amount. By single factor and orthogonal experiment, the optimum fermentation conditions of strain were as follows: the initial pH5.0, temperature of 35 ℃, culture time 48 h, inoculation amount 10%. Under optimal conditions, protopectinase activity was up to 4545.235 U/mL.
