摘要
大鼠亚硒酸钠2mg·kg^1·d^1 ip,连续2d,d 2给药后4 h ip顺铂5 mg/kg,能明显降低由ip顺铂引起的尿量、尿NAG活性及血尿素氮和血浆肌酐的升高,使尿渗透浓度维持正常水平,并使顺铂引起的大鼠肾脏近曲小管上皮细胞变性坏死明显减轻。亚硒酸钠在减少顺铂肾毒性的同时并不影响其抗癌效应。
The effect of sodium selenite on nephrotoxicity induced by cisplatin in rats was studied.Sodium selenite ip 2mg·kg-1·d-1 was injected for 2 d,and 4 h after ip sodium selenite on d 2 a single dose of cisplatine(5mg/kg)was injected ip.Selenite markedly reduced the elevation of urine flow,urinary N-acetyl-glucosaminidase(NAG),blood urea nitrogen and serum creatinine caused by cisplatin 5mg/kg and maintained urinary osmolarity at a normal level.The differences between the two groups are marked(p<0.01 or p<0.05).The histopathological impairment(the proximal tubule degeneration and necrosis)caused by cisplatin was alleviated significantly.The administration of sodium selenite 2 mg/kg ip to the tumor-bearing mice did not affect the antitumor activity of cisplatin.Cisplatin dissolved in normal saline which contained sodium selenite was injected ip to the tumor-bearing mice and the result showed that sodium selenite did not affect the antitumor activity of cisplatin.These results indicated that the protective effect of sodium selenite is probably due to the interaction between selenite and hydrolytic metabolites of cisplatin in the kidney.
