摘要
通过饲喂放线菌酮 ,提取小鼠肝脏的总RNA ,反转录合成cDNA第一链 ,利用RT PCR扩增出 110 0bp大小的片段 ,将其克隆入载体pBluescript,采取双链质粒直接测序 ,获得的序列与国外报道小鼠 2 5A合成酶基因序列具 99.8%的同源性。氨基酸序列分析表明 ,小鼠 2 5A合成酶与人2 5A合成酶具 6 8%同源性 ,并在其C端发现亲水基团和糖基化位点。
The 2 5 oligoadenylate system provides a universal response in mammals. 2 5A synthetase polymerizes ATP to a series of 2 5 oligoadenylate (2 5A). 2 5A activates a latent endoribonuclease (RNase L) which degrades viral RNA. In this paper the result of cloning and sequencing of the murine 2 5 oligoadenylate gene was reported. There are three different points between the nucleotide sequence of the cloned murine 2 5 oligoadenylate gene. The amino acid homology of murine 2 5A synthetase with human 2 5A synthetase was 68%. The C terminal portion of murine 2 5 A synthetase containes a hydrophilic region and a possible glycosylation site.
