摘要
为观察白介素1(IL-1)和白介素1受体拮抗剂(IL-1Ra)在人发育骨与软骨组织中的表达及分布,认识IL-1系统在人发育骨与软骨中的作用,用组织学及免疫组化技术,对临床经过石蜡包埋的人发育关节软骨组织进行定位定性检测,结果显示3月龄胚胎的软骨组织,IL-1β在肥大样改变的软骨细胞染色阳性。在4~6月龄胎儿,生长板软骨细胞自增殖后期到肥大样改变,IL-1β染色阳性。骨骺软骨(未钙化)内新生髓腔周围软骨细胞及骨髓腔内成骨细胞IL-1β也呈阳性染色。IL-1Ra蛋白表达区域基本类似于IL-1β,但表达的量较多,区域分布较广泛。结果显示,IL-1和IL-1Ra可能在人发育骨与软骨组织中通过旁分泌及自分泌方式协同发挥作用。
To investigate the expression and localization of interleukin - 1 receptor antagonist (IL- 1Ra) Protein and interleukin - 1 (IL- 1 ) in developing human long bones, and understand the function of IL-1 system in developing human long bones, developing human long bones were used to detect IL-a1Ra and IL-1βprotein expression by histology and immunohistochemistry. The result showed that IL-1Ra and IL- 1 protein were distributed in hypertrophic chondrocytes in cartilage of 3-month-old human embryo. In long bone of 4-6- month- old human fetus, IRAP and IL-1βProtein were distributed in the growth plate chondrocytes, neonatal marrow cavity chondrocytes in osteopiphysis and some osteoblasts; and IL-1Ra prortein expression was more numerous and broad distributing than IL-1β.the results suggested that IL-1Ra and IL-1β protein may play an important synergetic effect by both paracrine and autocrine mechanisms during endochondral bone development.
