组胺对脑细胞内游离钙浓度的影响及其机制

摘要 目的 观察组胺对脑细胞内游离钙浓度的影响 ,并初步探讨其作用机制。方法 利用荧光探针Fura - 2测定乳鼠脑细胞内游离钙的浓度。结果 组胺可明显升高脑细胞内游离钙浓度 ,而异丙嗪可以阻断组胺的这一效应。结论 组胺升高脑细胞内游离钙浓度是由H1受体介导的。 Objective To study the effect of histamine on free intracellular calcium in isolated neonatal rat brain cells and its possible mechanism. Method The Ca 2+ -sensitive fluorescence indicator Fura-2 was used. Results Histamine(0 1~ 0 3 mmol/L ) improved [Ca 2+ ]i by 37 17%,66 99% and 94 29%,respectively.This effect of histamine on [Ca 2+ ]i was completely antagonized by promethazine( 0 1 mmol/L ). Conclusion These results suggest that histamine can elevate [Ca 2+ ]i and may be relate to the brain ischemic injure,whereas promethazine can antagonize the increase of [Ca 2+ ]i induced by histamine.
