摘要
目的研究蕨麻提取物的抗艾滋病病毒Ⅰ型(HIV-1)活性及毒性。方法用HIV-1实验室适应株SF33、临床分离株XJDC257和020100968、假病毒颗粒9-14,18-36,74-2和Z20-11分别感染TZM-bl、MT-4,利用基于TZM-bl细胞的荧光素酶检测体系和基于MT-4细胞的P24抗原检测方法,观察蕨麻提取物抑制HIV-1病毒复制的活性;用不同稀释度的蕨麻提取物与TZM-bl.MT-4.PBMCs共培养,使用Cell Counting Kit-8检测活细胞的数量,观察蕨麻提取物对相应细胞的毒性作用。结果利用MT-4细胞和TZM-bl细胞测得蕨麻提取物对SF33的半数抑制浓度(IC50)及选择性指数(SI)分别为6.2μg/mL,26.4和4.7μg/mL,73.5。蕨麻提取物对HIV-1临床分离株XJDC257和020100968的IC50和治疗指数(TI)分别为2.1μg/mL,70.6和1.9μg/mL,77.6;对HIV-1假病毒颗粒9-14、18-36、74-2和Z20-11的IC50分别为1.8μg/mL、1.0μg/mL、3.4μg/mL和3.5μg/mL,TI分别为81.9、147.5、43.4和42.1。结论蕨麻提取物在体外具有一定的抑制HIV-1复制活性。
Objective To study the anti-HIV-1 activity and toxicity of the potentilla anserine L. extract. Meth- ods TZM-bl and MT-4 cell lines were infected with HIV-1 laboratory-adapted strain(SF33), two HIV1 clinical i- solates(XJDC257,020100968) and four HIV-1 pseudotype viruses(9-14,18-36,74-2 and Z20-11). The activity of in- hibiting replication of HIV-1 was observed respectively by using Luciferase assay system based on TZMbl cell line and P24 antigen expression detection based on MT-4 cell line.. The TZM-bl, MT-4 and PBMCs were co-cultured with potentilla anserine L. extract of different concentrations, then CCK-8 kit was used to detect the number of the living cells for evaluating the cytotoxicity of the extract. Results The ICs0 and SI of the potentill anserine L. ex- tract against SF33 was 6.2 /tg/mL, 26.4 by using MT-4 cell line and 4.7 Ixg/mL, 73.5 by using TZM-bl cell line. The ICs0 and TI of the extract against HIV-1 clinical isolates XJDC257 and 020100968 was 2.1 /lg/mL, 70.6 and 1.9 μg/mL, 77.6, respectively. The IC50 of the extract against the HIV-1 pseudotype viruses 9-14,18-36,74-2 and Z20-11 was 1.8 μg/mL, 1.0μg/mL, 3.4μg/mL and 3.5μg/mL,respectively and therapeutic index(TI) was 81.9, 147.5,43.4 and 42.1, respectively. Conclusion: The potentilla anserine L. extract could inhibit the replication of HIV-1 in vitro.
