不同实验条件对穿梭箱系统研究大、小鼠学习记忆行为学的影响 被引量:9

Effects of different experimental conditions on cognitive behavior of mice and rats for shuttle-box system

摘要 目的研究大、小鼠穿梭箱实验最佳训练条件,为神经系统相关实验研究提供参考。方法利用大、小鼠穿梭实验视频分析系统,考察不同实验条件对大、小鼠学习记忆形成及保持的影响。结果条件刺激时长为5 s时,KM鼠的单纯光刺激组学习成绩明显好于声光联合刺激组;单纯给予光刺激时,KM鼠条件刺激时长为10 s组的成绩明显好于5 s组;当给予声光联合刺激,刺激时长为10 s时,Wistar大鼠白天训练组与夜晚训练组的学习成绩并没有明显差异,但Wistar大鼠的学习成绩明显好于SD大鼠。训练达标后,KM鼠记忆至少能保持21 d;Wistar大鼠记忆至少保持28 d。结论大小鼠学习记忆的形成与实验动物的选择及实验条件的设置密切相关。适当延长条件刺激时间有利于KM鼠条件反射的形成;训练时辰对Wistar大鼠的记忆获得没有影响;无论是KM小鼠,还是Wistar大鼠,训练达标后,其记忆均能保持较长一段时间。 Aim To investigate the most suitable training conditions for experimental mice or rats and provide references for cognitive studies using shuttle-box system.Methods By shuttle-box video analysis system,the effects of different experimental conditions on the formation and retention of learning and memory behaviour of rats or mice were studied.Results For the conditioned stimulus duration 5 s,KM mice stimulated by single light showed a significantly better learning performance than those stimulated by the combination of sound and light.For the single light stimulus,with the conditioned stimulus duration 5 s and 10 s,the 10 s group performed significantly better than the 5 s group.For the training by sound and light combination stimulus,with the conditioned stimulus duration 10 s,Wistar rats showed a significantly better performance than SD rats,while there had no significant difference in learning performance between Wistar rats trained by day and those by night,respectively.After attaining the training standards,the memory in KM mice and Wistar rats could keep at least 21 days and 28 days,respectively.Conclusions The formation of learning and memory for rats or mice is closely related to the species of experimental animals and the experimental conditions.An appropriate extension of the conditioned stimulus duration is beneficial to the formation of the conditioned reflex of KM mice.Training time has no significant effect on the memory acquisition of Wistar rats.Once reaching the training standards,the learning and memory abilities of rats and mice will keep a longer time.
