雌三醇对人结肠癌细胞增殖的影响及其机理探讨

摘要 目的 探讨雌三醇 (E3)对人结肠癌细胞增殖的影响及其机理。方法 采用钙离子荧光指示剂 flour- 3/Am ,流式细胞仪动态检测 E3作用后 ,人结肠癌细胞内游离钙离子 [Ca2 + ]i水平的变化 ,同时用 MTT比色法测定细胞增殖情况。结果  1E3使细胞内 [Ca2 + ]i水平在 1分钟内下降 ,以后维持在一个较低水平 (2 7.88nmol/ L ) ,与静息状态细胞内[Ca2 + ]i水平 (94.4nmol/ L)相比有显著性差异 (P<0 .0 5 )。2 E3对癌细胞有促增殖作用 ,其浓度对数值与细胞增殖率呈显著性正相关 (r=0 .975 ,P<0 .0 5 ) ,其中 1× 10 - 5 mmol/ L浓度的 E3促增殖作用最大。结论  E3有促人结肠癌细胞增殖作用 ,但不引起细胞内 [Ca2 + ]i水平升高。 E3的促癌细胞增殖作用未见与经钙离子通道引起的非基因的快速调节作用有关 ,推测是基因调节作用的结果。 Objective To assess the effect of estriol on the proliferation of human colonic cancer cell lines and gain an insight into its mechanism.Methods Flow cytometry was used to continuously monitor the change of the intracellular free calcium[Ca 2+ ]i with calcium fluoresence indicator(fluo3/Am),meanwhile the MTT colorimetry was used to detect the proliferation of human colonic cancer cell lines(HT 29 cell).[WT5”HZ]Results [WT5”BZ]Firstly,estriol made the intracellular[Ca 2+ ]i level decrease in a minute,and then the intracellular[Ca 2+ ]i concentrations maintained a lower level (27.88 nmol/L),which was significantly lower than that (94.4nmol/L) of the intracellular[Ca 2+ ]i of static status cells ( P <0.05).Secondly,E 3 had the effect on improving the proliferation of HT-29 cells.The effect of E3(1×10 -5 mmol/L)on improving proliferation was great.There was a significant positive correlation between the cell proliferation rate and the logarithms of E3concentrations(r=0.975,P<0.05).Conclusion Estriol could promote proliferation of human colonic cancer cells,but it could not cause the increase of intracellular calcium level.The effect of estriol on promoting the proliferation of cancer cells is not related to non-genic rapid regulation by calcium-iron channel;it is supposed to be the result of gene-regulation.
