肿瘤坏死因子α和血管紧张素Ⅱ对人血管内皮细胞组织因子表达的影响 被引量:8

摘要 已知组织因子异常表达可启动血液凝固机制。用细胞发色底物法、RT-PCR等方法分别从蛋白活性、基因表 达水平研究与动脉粥样硬化及高血压发病相关的肿瘤坏死因子、血管紧张素Ⅱ对血管内皮细胞组织因子表达的影响。 结果表明:肿瘤坏死因子α和血管紧张素Ⅱ均诱导血管内皮细胞组织因子的表达,并使其mRNA表达增加。此项研 究为进一步探讨动脉粥样硬化和高血压的共同发病机制打下基础。 Cell chromogenic substrate assay and RT-PCR method were applied to study the effects of tumor necrosis factor a and angiotensin Ⅱ on the expression of protein activity and mRNA of tissue factor. ECV304 endothelial cells were stimulated with tumor necrosis factor a and angiotensin Ⅱ. Results showed that tumor necrosis factor a and angiotenisn Ⅱ induced tissue factor activity. Tissue factor activity on ECV304 cell surface increased significntly after stimulated with 100U/mL tumor necrosis factor a for 6h (P<0.05 ) or with 10^-6 mol/L angiotensin Ⅱ for 12h (P<0.05 ). Moreover,the expression of TF mRNA increased compared with the control group. Results is of benefit to the further research on the common mechanism of atherosclerosis and hypertension.
