聚合酶链反应凝胶电泳的摄影

摘要 为提高聚合酶链反应凝胶电泳的图像质量 ,用不同的胶卷 ,设定不同的曝光和冲洗条件 ,对凝胶电泳进行摄影条件摸索。结果 :(1)对弱荧光带拍摄 ,ISO10 0 / 2 1°黑白胶卷曝光条件为 :f/ 2 ,时间 30~ 6 0秒 ;ISO40 0 / 2 7°黑白胶卷曝光条件为 :f/ 2 .8,时间 5秒左右。 (2 )对强荧光带拍摄 ,ISO10 0 / 2 1°黑白胶卷曝光条件为 :f/ 2或 f/ 2 .8,时间 10~ 30秒 ;ISO40 0 /2 7°黑白胶卷曝光条件为 :f/ 4,时间 1~ 4秒。冲洗可用原液浓度 D- 72显影液。照片图像效果满意。提示 :电泳荧光亮度和正确曝光是图像质量的关键。 In order to improve the image quality of the polymerase chain reaction,the photographic conditions of gel dectrophoresis were studied with different film and different exposuring and developing condition The resules showed that(1)for the light fluorescent band,exposuring conditions of the ISO100/21° black white film are f/2 and 30-60 seconds,and the ISO400/27° black-white film f/2 8 and about 5 second,(2)for the strong fluorescent band,the ISO100/21°black-white film f/2 or f/2 8 and 10-30 seconds,and the ISO400/27° blackwhite f/4 and 1-4 seconds The D-72 developer of the primary concentration were used to develop The effect of photograph is satisfactory It suggested that the fluorescent brightness and exact exposure to light is of importance for image quality
