右美托咪定对渥曼青霉素引起的海马CA1区神经元凋亡的影响 被引量:4

摘要 目的探讨右美托咪定(Dex)对渥曼青霉素(Wort)引起的海马CA1区神经元凋亡的影响。方法以SD大鼠胎鼠海马CA1区神经元为研究对象,(1)Wort对海马CA1区神经元的致凋亡作用:将SD大鼠海马CA1区神经元随机分为4组,对照组及Wort不同剂量组(Wort 0.3、1.0、3.0组),对照组不作处理,Wort 0.3、1.0、3.0组分别接受0.3、1.0、3.0μmol/L Wort处理24 h;(2)Dex的抗凋亡作用:将SD大鼠海马CA1区神经元随机分为5组,对照区(不作处理)、Wort组(3μmol/L Wort处理24 h)、Wort+Dex 0.1组(3μmol/L Wort+0.1μmol/L Dex处理24 h),Wort+Dex 1.0组(3μmol/L Wort+1.0μmol/L Dex处理24 h)和Wort+Dex 10.0组(3μmol/L Wort+10.0μmol/L右美托米啶处理24 h)。采用DAPI荧光染色法法计数细胞凋亡数,计算凋亡率。结果 3个剂量的Wort均可引起海马CA1区神经元凋亡,以Wort 3.0组最为明显;Wort+Dex 0.1、1.0、10.0组与Wort组比较细胞凋亡率明显下降(P<0.05,P<0.01),但仍明显高于对照组(P<0.01)。结论 Dex能有效抑制Wort引起的海马CA1区神经元凋亡,具有神经保护作用。
