摘要
目的 :观察三氧化二砷 (As2 O3)对人膀胱癌细胞的生长抑制作用 ,并探讨其机制。方法 :采用 MTT法检测不同浓度的 As2 O3对人膀胱癌细胞株 BIU- 87的生长抑制率 ,原位末端转移酶标记技术检测细胞凋亡 ,SABC免疫组织化分析 BIU- 87细胞中 bcl- 2的表达。结果 :As2 O3可有效地抑制 BIU- 87细胞生长 ,具有时间、浓度依赖性特点 ;经药物作用后 ,膀胱癌凋亡细胞明显增多 ,凋亡率随作用时间的延长而增高 ,BIU- 87细胞中的bcl- 2表达显著降低。结论 :As2 O3可显著抑制膀胱癌细胞生长 ;通过下调 bcl- 2基因表达诱导细胞凋亡是其作用机制之一。
Purpose: To observe the effects of arsenic trioxide (As 2O 3) on the growth of human bladder cancer cells and illustrate its mechanisms. Methods: The growth inhibition rates of human bladder cancer cell line BIU 87 by various concentration As 2O 3 were studied by MTT method, cell apoptosis of BIU 87 cells was observed by SABC immunohistochemical method.Results: As 2O 3 could inhibit the growth of BIU 87 effectively with the increase of apoptosis cells, which depended on the action time and concentration of the drug . Bcl 2 expression of BIU 87 cells was decreased significantly.Conclusions: As 2O 3 could significantly inhibit the growth of bladder cancer cells. Inducing cell apoptosis by down regulating the expression of bcl 2 gene might be one of its action mechanisms .
