取材时期对文冠果优树芽启动的影响 被引量:5

Effect of Collection Time of Explants on Bud Sprouting in Superior Tree of Xanthoceras sobifolia

摘要 将3个不同取材时间(5月10日、6月10日和7月10日)文冠果芽外植体接种在含有不同植物生长调节剂(BA、IBA、NAA)组合的MS培养基中培养,考查取材时间和植物生长调节剂对芽萌发、生长及其生根的影响。结果显示:3个时期中,5月份取材的顶芽萌发率和生长最好,7月份最差,几乎没有腋芽萌发;5月份取材外植体在BA0.1 mg/L条件下芽的萌发和生长较好,芽萌发率和芽高分别为88.4%和2.5 cm。6月份取材的外植体在MS+BA0.5 mg/L+NAA0.2 mg/L条件下,芽的萌发率比其他植物生长调节剂条件下的高,约51.3%,萌发芽高为1.5cm;5月和6月份取材获得的芽继代不好,容易死亡,在MS+BA0.5 mg/L+NAA0.2 mg/L培养基条件下继代最好,其4周的存活率为24%;芽生根的最佳培养条件为1/2MS+IBA0.8 mg/L+NAA0.4 mg/L。 Bud explants of Xanthoceras sorbifolia Bunge cut from mother trees on three different days(May 10th,June 10th,and July 10th) in 2010 were inoculated on MS media with different plant growth regulators(BA,IBA,and NAA).The effects of collection time of the explants and the combination of plant growth regulators on bud sprouting,growth and rooting were studied.Results showed that the sprouting and growth of buds obtained on May 10th were better,while those collected on July 10th were worst,and almost no buds sprouted.The optimal condition for sprouting and growth of buds inoculated on May 10th was MS medium supplemented with 0.1 mg/L BA,with a bud sprouting rate of 88.4% and an average shoot height of 2.5 cm.The sprouting rate of the buds collected on June 10th inoculated on MS medium supplemented with 0.5 mg/L BA and 0.2 mg/L NAA was 51.3%,and the average shoot height was 1.5 cm.The subculture of shoots obtained from the buds collected on May 10th or June 10th easily died,and the survival rate of shoots four weeks after the subculture was 24% under the subculture condition of MS medium containing 0.5 mg/L BA and 0.2 mg/L NAA.The optimal condition for shoot rooting was 1/2 MS medium supplemented with 0.8 mg/L IBA and 0.4 mg/L NAA.
