摘要
目的:研究异氟醚麻醉对大鼠脑内一氧化氮合酶(NOS)活性的影响。方法:20 只雄性SD大鼠,随机分为4 组(每组n= 5 只),分别吸入0220m in(对照组)、0.8% 异氟醚20m in、1.5% 异氟醚20 分、1.5% 异氟醚1h。然后断头取脑,用分光光度法测定大鼠大脑皮层、小脑、脑干NOS活性。结果:异氟醚以剂量依赖方式抑制小脑NOS活性。0.8% 、1.5% 异氟醚均显著抑制大脑皮层、脑干NOS活性,但两个浓度异氟醚所产生的效应无显著差异(P> 0.05)。1.5% 异氟醚麻醉20m in 与1h,对大鼠各脑区NOS活性的影响无显著性差异(P> 0.05)。结论:异氟醚麻醉抑制脑NOS活性,但NOS活性变化与麻醉药浓度有关,且存在脑区域上的差异。
Objective:To investigate the influence of isoflurane anesthesia on nitric oxide synthase(nos)activity in different rat brain regions.Methods:20 male SD rats were randomly divided into four groups.Rats were unanesthetized(as control) or anesthetized with 0.8% isoflurane for 20 minutes、1.5% isoflurane for 20 minutes、1.5% isoflurane for one hour,respectively.Then rats were decapitated and cerebral cortex、cerebellum and brain stem quickly harvested and homogenized.Brain extracts were then examined for NOS activity.NOS activity was determined by the hemoglobintrapping.binding of NO to oxyhemoglobin produces methemoglobin and nitrate.The change in methemoglobin concentration is measured spectrophotometrically to estimate NOS activity.Result:Isoflurane dose-dependently inhibited the NOS activity in rat cerebellum(P<0.01).At 0.8%、1.5% isoflurane,NOS activity was significantly inhabited in conted and brain stem(P<0.01 versus control group)while there was no difference between the effects at the two doses(P>0.05).At1.5% isoflurane,NOS activity in all researched brain regions was not significantly altered when anesthetic time prolonged from 20 minutes to one hour(P>0.05).Conclusions:Isoflurane can inhibit brain NOS activity. The effect of isoflurane appear to vary in different brain regions and at different concertrations.
