摘要
目的 探讨FasL及pRb在视网膜母细胞瘤中的表达及意义。方法 用免疫组织化学SP方法检测45例视网膜母细胞瘤及12例正常视网膜组织中FasL及pRb蛋白的表达。结果 视网膜母细胞瘤组织表达pRb,比正常组织显著降低(P=00009)。视网膜母细胞瘤和正常组织均高表达FasL。结论 FasL的表达是贯穿视网膜母细胞瘤发生及发展过程中的一个重要事件,pRb低表达与肿瘤的发生有密切关系。
ObjectiveTo investigate the significance of FasL and pRb expression in retinoblastoma.MethodsAll materials were archival formalin fixed and paraffin embedded samples.Both FasL and pRb in 45 cases of retinoblastoma and 12 cases of normal retina were investigated.The biotinylated second antibody and enzyme conjugated immunohistochemical staining technique was used.ResultsThe expression of pRb in retinoblastoma and normal retina were 1.778±0 876 and 2 833±1 115,respectively( P =0 0009),immunostaining for pRb in normal retina were significantly higher than that in the retinoblastoma.The expression of FasL in retinoblastoma and normal retina were 3 021±0 847 and 3 583±0 669,respectively( P =0 023).Normal retina tissue stained very strongly positive for FasL and the retinoblastoma stained slightly weaker than the normal retina tissue,exhibiting FasL was high regulation in normal retina and retinoblastoma.ConclusionHigh regulation of expression of FasL in retinoblastoma and normal retina suggested that it is a throughout event which implicated a novel mechanism immune escape.The decrease of pRb expression in retinoblastoma is closely related to tumor pathogenesis.
