摘要
目的 研究大鼠角膜移植排斥反应发生机理。方法 制作模型(Wistar大鼠18只,豚鼠9只)。观察角膜透明度、新生血管。角膜和外周血行免疫检测。并对角膜进行扫描电镜观察。结果 术后1周,植床出现新生血管、角膜水肿、基质增厚;术后1月,植床瘢痕化,但水肿减轻。免疫组化显示:移植后3天,血中Th/i细胞IL2R+细胞升高,15天时角膜植片内有以TC/S细胞浸润为主的淋巴细胞。结论 大鼠穿透性角膜移植术后排斥反应有着与机体其它器官相似的移植排斥过程。
ObjectiveTo investigate the rejection mechanisms of corneal penetrating xenograft.MethodsThe corneal penetrating xenograft models were made in 18 Wistar rats and*'9 guinea pigs.The diameter of graft was 6 0*!mm,and diameter of receipt bed was 0 5*!mm shorter than the graft.The graft was continuous sutured by nylon wire.Corneal pellucidity,edema and neovascularization were observed by opton slit lamp microscopy every 4*!d,and stopped till 30*!d after keratoplasty.Rejection value (RV) was calculated according to Holland’s grade.On 6,10*!d (one eyeball)and 15*!d(two eyeballs) after keratoplasty,immunohistochemical study was performed in the cornea.On 3*!d preoperation and 3,9,15,21,30*!d postoperation,1 5*!mm blood was gotten out from the rat’s hearts,and the changes of lymphocyte subgroup were observed.On 12,20*!d after keratoplasty,the corneal constitution and form of the animals were respectively observed and taken a photograph by scanning electron microscope.Immunological study was performed in the cornea and the peripheral blood.ResultsIn the first week of postoperation,neovascularization,corneal edema and thickness of stroma occurred in recipient site.In the first postoperative month,the recipient site cicatrized and corneal edema lessened.Immunohistochemical studies showed that Th/i cells and IL 2 R + cells were increased in the peripheral blood on 3*!d after keratoplasty,At 15*!d after keratoplasty,the major infiltrating cells were Tc/s cells in the graft corneas.ConclusionThe rejection of corneal penetrating xenograft in rats may be similar to that of other organs.
