呼吸道合胞病毒和人巨细胞病毒混合感染对感染细胞的影响 被引量:2

摘要 目的 探讨呼吸道合胞病毒 (RSV)和人巨细胞病毒 (HCMV)混合感染对感染细胞的影响。方法 用四甲基偶氮唑盐 (MTT)染色方法检测下列四组标本的吸光度A值 (曾称光密度OD) ,包括 :未感染病毒细胞组、RSV感染细胞组、RSV和HCMV同时混合感染细胞组、HCMV感染 12h后再感染RSV细胞组 (每组 2 0孔 ,共 80孔 ) ;流式细胞分析仪检测下列六组标本细胞分裂周期 ,包括 :正常细胞组、RSV感染组细胞、HCMV感染组、HCMV和RSV同时感染细胞组、RSV感染 12h后再感染HCMV细胞组、HCMV感染 12h后再感染RSV细胞组 (每组 12孔 ,共 72孔 )。结果  (1)未感染病毒组A值(1.5 0± 0 .12 )明显高于RSV感染组 (1.12± 0 .13)和混合感染组 (1.0 9± 0 .0 8) (P <0 .0 1) ,两种病毒混合感染与RSV单独感染组A值差异无显著性 (P >0 .0 5 )。 (2 )正常细胞组S期细胞比例 (15 .31±1.0 8)较病毒感染组为高 (P <0 .0 1) ,RSV单独感染组 (10 .6 5± 0 .72 )较HCMV单独感染组 (5 .6 5±1.35 )及RSV和HCMV混合感染组S期细胞比例 (5 .0 2± 1.0 8、4.79± 0 .2 8、5 .14± 0 .5 6 )为高 ,HCMV单独感染与混合感染组S期细胞所占比率差异无显著性 (P >0 .0 5 )。结论 RSV单独感染和与HCMV混合感染 ,二者对感染细胞活细胞数影响基本相同 ; Objective To investigate the damage to cultured cells caused by respiratory syncytial virus (RSV) and human cytomegalovirus (HCMV) coinfection. Method MTT colorimetric assay was used to measure the A value of the following four WI 38 cell samples, including cells which were not infected with viruses, infected with RSV, infected with RSV and HCMV simultaneously, cells coinfected with HCMV 12 hours after RSV infection, there were altogether 80 samples, 20 samples per group. The cell cycle progression of the following six WI 38 cell samples were investigated by using flow cytometry, including the normal cells, cells infected with RSV, cells infected with HCMV, cells coinfected with RSV and HCMV simultaneously, cells coinfected with HCMV 12 hours after RSV infection and cells coinfected with RSV 12 hours after HCMV infection, there were altogether 72 samples, 12 samples per group. Results (1) The A value of normal cell samples (1.50±0.12) was greater than that of cells infected with RSV (1.12±0.13) and coinfected with RSV and HCMV (1.09±0.08) ( P < 0.01). The A value of RSV infected cells was not significantly different from that of RSV and HCMV coinfected cells ( P > 0.05). (2) The proportion of S phase cell of normal cells (15.31±1.08) was higher than that of cells infected with virus ( P <0.01), and the proportion of S phase cell of RSV infected cells (10.65±0.72) was higher than that of HCMV infected cells (5.65±1.35) and RSV and HCMV coinfected cells (5.02±1.08, 4.79±0.28, 5.14± 0.56) ( P <0.01). No significant difference was found in the proportion between HCMV infected cells and RSV and HCMV coinfected cells ( P >0.05). Conclusion The numbers of living cells were not different between RSV infected cells and RSV and HCMV coinfection. The cell cycle was inhibited by HCMV and RSV infection, but the inhibition induced by single HCMV infection and coinfection with RSV and HCMV was stronger than that by single RSV infection.
