摘要 目的转染Pokemon表达载体到MCF-7细胞中,探究Pokemon对乳腺癌细胞增殖的影响。方法构建Pokemon基因表达质粒,瞬时转染Pokemon表达质粒到乳腺癌细胞中,Western blot检测pokemon的蛋白水平表达情况,通过MTT和平板克隆形成试验法观察Pokemon对乳腺癌细胞增殖及克隆形成能力的影响。结果MTT法显示,随着培养时间的增加,MCF-7细胞在转染Pokemon后的生长速度明显增加;细胞平板克隆形成试验显示,MCF-7细胞在转染Pokemon后克隆数明显增加,由此可以说明Pokemon能增强癌细胞MCF-7克隆形成能力。结论Pokemon作为一个致癌基因,能促进乳腺癌MCF-7细胞系的增殖并增加其克隆形成能力。 Objective To assess the effect of Pokemon gene over-expression on the proliferation ability of MCF-7 cells by transfection of Pokemon expression vector into the MCF-7 cells. Methods We trans- fected the constructed Pokemon expression plasmid into the MCF-7 cells. We detected the Pokemon ex- pression by Western blot, and observed the proliferation of MCF-7cells by MTF. Plate efficiency assay wasapplied to analyze the clone formation activity of MCF-7. Results As the culture time increased, the growth rate of MCF-7 cells after tmnsfected with Pokemon expression plasmids was increased significantly. The increased number of cloning of MCF-7 cells with Pokemon expression plasmid compared with the control cells indicated that Pokemon could boost the clone fomkafion ability of MCF-7 cells. Conclusions As an oncogene, Pokemon could increase the proliferation ability and colony formation abihty of MCF-7cells.
