摘要
目的探讨KiSS-1基因转染对骨肉瘤MG-63细胞株增殖的影响及其可能机制,为骨肉瘤的基因治疗提供实验室依据。方法将KiSS-1基因转染至骨肉瘤MG-63细胞株,应用免疫组化法检测MG-63-KiSS-1蛋白表达,CCK-8法检测细胞增殖情况,流式细胞术检测各期细胞比例。结果 MG-63-KiSS-1细胞KiSS-1蛋白高表达、增殖明显减慢、G2/M期细胞比例明显增高。结论 KiSS-1基因转染MG-63细胞后可抑制其增殖,其机制可能与阻滞细胞周期于G2/M期有关。
Objective To investigate the influence of KiSS-1 transfection on the proliferation of osteosarcoma cell,and to explore the feasibility of gene therapy for osteosarcoma.Methods A eukaryotic expression vector carrying the KiSS-1 gene(rAAV-KiSS-1)was transduced into MG-63 cells with Lipofectamine2000;Immunohistochemistry was applied to analyse KiSS-1 expression of MG-63-KiSS-1 cells;CCK-8 was used to examine cell proliferation and Flow cytometry was used to analyse the cell percentage of each cycle.Results MG-63-KiSS-1 cells highly expressed KiSS-1 protein.The cell proliferation was slow down obviously and the cell percentage of G2/M increased after rAAV-KiSS-1 infection.Conclusion The transfection of KiSS-1 gene inhibits the MG-63 cells proliferation,and the mechanism may be relate to blocking the cell cycle at G2/M period.
