摘要
目的:探讨清肺口服液含药血清对呼吸道合胞病毒(RSV)感染的人胚肺成纤维细胞(HELF)细胞间黏附分子(ICAM-1)、白细胞介素-8(IL-8)基因表达的影响。方法:RSV攻击体外培养的HELF细胞后,以清肺口服液含药血清,利巴韦林含药血清及空白血清干预,采用实时荧光定量PCR法(RealTime-PCR)测定各组人胚肺成纤维细胞ICAM-1、IL-8 mRNA表达变化。结果:病毒感染后ICAM-1、IL-8以GAPDH为内参的△Ct值明显小于正常细胞组及生理盐水血清对照组,而清肺口服液含药血清、利巴韦林含药血清作用后△Ct值显著增大。结论:(1)RSV病毒作用后ICAM-1及IL-8 mRNA表达水平显著升高;(2)清肺口服液可以降低ICAM-1、IL-8的表达,缓解RSV感染后细胞的异常变化,起到间接预防RSV肺炎的作用。
Objective : To investigate the effects of Qingfei oral liquid on expressions of Interleukin (IL) - 6 and Intedeukin (IL) -8 in human epithelial cells( Hep-2) infected with respiratory syncytial virus(RSV). Methods:Hep- 2 cells were intervened with serum containing Qingfei oral liquid, Ribavirin and blank serum, respectively. In each of these three groups, intervention was conducted before, simuitaneously or after RSV infection ( I , II, m ). Expression levels of IL - 6 and IL - 8 were detected by enzyme linked immunosorbent assay (ELISA)after collecting the cellular supernatant. Results:After 48 and 72 hours of cultiva- tion, expression levels of IL - 6 and IL - 8 in virus control group were significantly higher than those in Qingfei oral liquid groups ( I , II, III ) or Ribavirin group( II, III ). No significant difference was found between blank serum control groups( II, III ) and the virus control group. Difference between the Qingfei oral liquid groups ( I , II, III )and the Ribavirin groups ( II, III )was insignificant either. Expression groups ( I , II , III) were slightly different after 48 or 72 hours of cultivation. Conclusion:Qingfei oral liquid can decrease the over - expressions of IL - 6 and IL - 8 so as to relieve inflammation. Furthermore, better effects can be achieved when it was administrated in early stage of RSV infection. These foundings may be related to the antiviral mechanism of Qingfei oral liquid.
