自体嗅粘膜联合β-七叶皂甙钠对脊髓损伤后FasL及Caspase-3表达的影响

Effects of autologous olfactory mucoma combined with β-aesine sodium on expression of FasL and Caspase-3 after spinal injury

摘要 目的探讨自体嗅粘膜联合β-七叶皂甙钠对Caspase-3及FasL表达与神经再生功能的影响。方法 SD大鼠随机分为损伤对照组(A组),自体嗅粘膜移植组(B组),自体嗅粘膜联合β-七叶皂甙钠组(C组),β-七叶皂甙钠组(D组)。每组分为1、3、7、14 d 4个时间点。制作脊髓半切模型,观察组织病理学改变;对动物后肢功能进行BBB评分;采用免疫组化法检测FasL、Caspase-3的表达。结果脊髓损伤后B、C、D组BBB评分均高于A组,FasL、Caspase-3阳性细胞数均较A组少,C组BBB评分较B、D组高,FasL、Caspase-3阳性细胞数较B、D组少。B、D组间无显著性差异。结论自体嗅粘膜与β-七叶皂甙钠两者之间有协同作用,可明显改善神经运动功能;此种保护作用可能与联合治疗能够抑制FasL、Caspase-3途径的细胞凋亡有关。 Objective To approach the influence of autologous olfactory mucoma combined with β-aesine sodium on expression of FasL and Caspase-3 in injuried spinal cord. Methods Sprague-Dawley rats with spinal T10 hemisection were divided into four groups as follows: control group(group A),autologous olfactory mucoma transplantation group(group B),combination group(group C) and β-aesine sodium group(group D).Each group was subdivided into four groups: 1,3,7 and 14d.for evaluating pathological changes of the spinal cord,the Basso-Beattie-Bresnahan(BBB) scales of hindlimb movement function were carried out.The expression of FasL and Caspase-3 in injured spinal cord were detected by immunohistochemistry analysis.Results BBB scores of group B,C and D were higher than group A.FasL and Caspase-3 positive cells in group B,C and D were less than group A.BBB scores of group C were higher than group B and D,and FasL and Caspase-3 positive cells of group C were less than group B and D.There was no significant difference between group B and D.Conclusion Synergistic effects occurs for promoting the recovery of hindlimb motor function after spinal injury combined autologous olfactory mucoma with β-aesine sodium,which also help to restrain apoptosis by reducing expression level of FasL and Caspase-3.
