金黄扶正茶对免疫抑制小鼠细胞免疫功能的影响 被引量:7

Influences of Jinhuang Fuzheng Tea on Cellular Immune Function in Immunosuppressed Mice

摘要 目的:探讨金黄扶正茶对免疫抑制小鼠脾脏T、B淋巴细胞增殖及腹腔巨噬细胞免疫功能的影响。方法:金黄扶正茶高、中、低剂量组分别连续给昆明种小鼠ig 10 d,同时隔日sc注射环磷酰胺(30 mg·kg-1)造成免疫抑制模型。采用四甲基偶氮唑蓝(MTT)法测定刀豆蛋白A(Con A)诱导脾脏T淋巴细胞增殖和脂多糖(LPS)诱导B淋巴细胞增殖的影响及自然杀伤细胞(NK细胞)和淋巴因子激活的杀伤细胞(LAK细胞)活性,酶法检测腹腔巨噬细胞分泌一氧化氮合酶(NOS)的活性,用全自动酶标仪检测巨噬细胞吞噬中性红能力。结果:金黄扶正茶低、中、高剂量组均能显著提高免疫抑制小鼠的T,B淋巴细胞的吸光度(A,P<0.01);而中、高剂量组能显著提高免疫抑制小鼠NK细胞、LAK细胞活性、腹腔巨噬细胞吞噬活性及诱导型一氧化氮合成酶(iNOS)活力(P<0.01或P<0.05)。结论:金黄扶正茶能促进免疫抑制小鼠脾淋巴细胞增殖,增强NK细胞、LAK细胞活性,提高免疫抑制小鼠腹腔巨噬细胞吞噬活性,从而增强免疫抑制小鼠的细胞免疫功能。 Objective:To observe the effects of Jinhuang Fuzheng tea on spleen T,B lymphocyte proliferation and peritoneal macrophage immune function in the Immunosuppressed mice.Method: Mice were ig administrated Jinhuang fuzheng tea at high,middle and low dose for 10 days.The immunosuppression mouse model was caused by subcutaneous injection of cyclophosphamide(30 mg·kg-1) every other day.The determination of concanavalin A(Con A) induced T lymphocyte proliferation and lipopolysaccharide(LPS) induced B lymphocyte proliferation and natural killer cells(NK cells) and lymphokine-activated killer cells(LAK cells) activity was examined by tetrazolium blue(MTT).The inducible nitric oxide synthase(iNOS) secreted by peritoneal macrophages activity was measured by enzymic method.Automatic detection microplate reader was used to determine the capability of phagocytosis neutral red.Result: The immunosuppressed mice which were given Jinhuang Fuzheng tea at low,medium and high dose groups showed a increased T and B lymphocyte A value(P0.01);The medium and high dose groups significantly improved NK cells LAK cell activity,phagocytic activity of peritoneal macrophages and iNOS activity in immunosuppressive mice(P0.01,P0.05).Conclusion: The Jinhuang Fuzheng tea can promote the immune function in Immunosuppressed mice.
