miRNA在结直肠癌发生发展过程中的调控机制 被引量:6

Mechanism of miRNA in regulating carcinogenesis of colorectal cancer

摘要 内源的功能性非编码小分子RNA-miRNA,在转录后水平调控目的基因的表达,进而参与调节细胞周期、凋亡、分化、发育以及肿瘤发生等多种生物学过程。miRNA能够调节DNA甲基化修饰,并对许多结直肠癌相关重要信号通路具有调控作用。miRNA表达异常会引起WNT、EGFR通路过度激活,TGFβ信号响应缺失,以及p53等抑癌基因的失活,导致结直肠癌发生。miRNA的甲基化修饰及其与靶基因的单核苷酸多态性(SNP)是miRNA调控结直肠癌发生的重要分子机理。本文总结了miRNA在结直肠癌发生发展过程中的调控作用及其分子机制,并初步探讨了miRNA表达异常致使肿瘤发生的影响因素。
