LAK—88/DC细胞系是将两只正常雄性615近交系小鼠的脾细胞混合,经ConA制激后,分离淋巴细胞。于含有3u/ml (?)—2的RPMI1640全培养基中培养并连续传代而建成。它是一株IL-2依赖、Thy—1.2^+的T淋巴细胞系,核型为40、xy。LAK—88/DC呈悬浮培养、生长迅速,迄今已传至160余代。支原体检测阴性,已用于IL-2制剂的活性检测。
A cell line designated as 'LAK-88/DC' has been established from mixed spleen cell culture of two normal male 615 inbred mice (H- 2~k), through ConA (10 μg /ml ) activation, lymphoblast isolation, making suspension culture and successive passage in RPMI 1640 medium with 20% FCS and 3u/ml of rIL-2, under37℃, 5 %CO_2 condition. It has been already successfully passed for more than 130 generations. 'LAK-88/DC' is a rapid growing suspension cell line It multiplied approximately 30.25 folds in number within 6 days with mitotic index reaching 2.1 % on the 4th day. 'LAK- 88/DC' s an IL-2 dependent and Thy- l, 2^+ diploid T-lymphoblast cell line of 615 mice. Its karyotype is 40 XY, no marker chromosome has been observed. In the 0.5-1.0u/ml rIL -2 and 15% FCS RPMI 1640 medium its growth might be optimal, but with higher concentration of IL-2 (>3 u /ml ) and FCS (20%) it grew even not so well. This cell line is the first IL-2-dependent T-lymphoblast cell line established from 615 inbred strain mice in China. It was successfully used for detecting the biological activities of IL- 2 products and aboptive immuno- prophylaxis and immunotherapy of L615 leukemia. It could be preserved in liquid nitrogen and its resuscitation although was not so easy but still possible. PPLO were not detected. This cell line might be of great value for practical use.
