纤维素内切酶Ⅲ再折叠过程中存在中间体(英文)

Existence of an intermediate in the refolding process of EndoglucanasesⅢ

摘要 本文研究了尿素变性的纤维素内切酶Ⅲ的再折叠.通过更灵敏的方法—相图法,本研究发现在2 mol/L~4 mol/L尿素间,纤维素内切酶Ⅲ的再折叠过程中存在至少一个中间体,聚沉曲线也进一步验证了此中间体的存在.该中间体的存在个数可进一步通过蛋白动力学曲线得以确定. Protein refolding is one of the most perplexing problems in molecular biology. The protein refolding process of endoglucanases IT was studied in this paper. It was found that there is a transition state between 2 mol/L and 4 mol/L urea during its refolding through a more sensitive method phase diagram, which is different from the results Usha Arunachalam had ever reported. The existence of the transition state has been confirmed through aggregation measurements. Future study will focus on the number of the intermediates through kinetic study.
