摘要
目的对小样本量骨结核临床分离菌株基因型的特点进行初步研究,以了解引起骨结核与肺结核的病原菌在基因型方面有无差异,为探讨两者之间的关系提供相关依据。方法利用间隔区寡核苷酸分型法(Spoligotyping)和数目可变串联重复序列分析(VNTR)分型法对骨结核与肺结核的临床分离菌株进行分型,从而确定每株结核分枝杆菌的基因型。结果Spoligotyping分型方法确定了在14株肺结核临床分离菌株中有9株为Beijing基因型,2株为T2基因型,另外3株分别为MANU2型、Haaslem3型以及未知型,VNTR可将14株肺结核临床分离菌株完全区分为14个基因型。而对于14株骨结核临床分离菌株来说,Spoligotyping和VNTR两种分型方法的结果均显示其具有完全相同的基因型。结论骨结核与肺结核临床分离菌株基因型差异巨大。特定型别(Spoligotyping:777777777777771,VNTR:013582202252)的结核分枝杆菌在骨结核流行菌株中占据主要地位,较之其他型别更易引起骨组织的病蛮。
Objective We used small size samples of Mycobactenum tuberculosis getting from bone tuberculosis and pulmonary tuberculosis patients for research in order to understand the differences of genotype between them. It will contribute to offer some relevance proofs for working out the relationship between bone tuberculosis and pulmonary tuberculosis. Methods Spoligotyping and VNTR methods were applied to 14 clinical isolated strains getting from bone tuberculosis patients (B-MTB) and 14 strains from pulmonary tuberculosis patients (P-MTB). By this, we can make sure the genotype of all Mycobactezium tuberculosis strains. Results 14 P-MTB isolates can be divided into 5 patterns by Spoligotyping, including 9 Beijing genotype strains, 2 T2 genotype strains and 3 other genotypes, and 14 patterns by VNTR. In contrast, 14 B-MTB were grouped into the completely same cluster assessed by both methods. The cluster can be shown as a column of figures 777777777777771 by Spoligotyping and 013582202252 by 12 VNTR locus we used. Conclusions There are significant differences in genotype of MTB clinical isolates strains getting from bone tuberculosis patients and pulmonary tuberculosis ones. In B-MTB, there are none of Beijing genotype strains. Furthermore, all of B-MTB possess the same gen- type (Spoligotyping:777777777777771, VNTR:013582202252). So, this particular genotype strains may action as the main epidemic strains of bone tuberculosis, and it can be able to invasion bone tissues much easier than other strains.
